Neo Mr. Do! is worth playing at your local arcade, if you can find it. But despite all the glitter, eccentricities, idiosyncracies, and what have you, the original is still the better game.
The strange thing is that Mr. Do hasn't really changed much in 14 years. The graphics are a little better, but the levels and game play haven't changed much since I was playing this on my friends CollecoVision. The control is basically the same as before, and the enemies (especially their AI) hasn't improved at all. You can build up your characters attack, but you can just as easily dodge the enemies. Not much of a challenge.
Playing Neo Mr. Do! might be something like playing Dig Dug while on an acid trip. If that's the kind of experience you're looking for, this game is just perfect for you. But if you want a more rational, logical, and non-insane experience, you should avoid this game at all costs.
