Combo Crew is a beat 'em up game where a character needs to fight waves of enemies in small, single-screen arenas. There are two game modes. King of the Tower is the story campaign where stages need to be completed grouped together in different environments such as 'warm up', 'the cell' and 'dojo' to defeat a final boss. The initial stages also serve as a tutorial. The second one is called Combo Crew and it consists of different rounds where specific challenges need to be completed.

The character's movement is not controlled, the player only controls the attacks and their direction through taps and swipes. There are two modes of control, either with one hand, or two hands where only the thumbs are used. By swiping with one finger left or right a single attack is performed. The game however encourages you to perform combos and to string them together. This is done by swiping up, down, left or right with two fingers. The character will then automatically run towards the nearest enemy in that direction and perform the combo. Certain characters can block and these need to be taken out with a charged attack by holding down one finger. Enemies' incoming attacks and incoming projectiles are blocked by tapping once, this can also be done during a combo. While most combos are performed automatically, there are juggles where an enemy is launched in the air. An air attack can then be performed by swiping once more. After performing well a super can be activated, where swiping enemies multiple times unleashes a fury of hits in all directions. Enemies can also be stunned or be launched against a wall, hitting all enemies on the way.

Initially, each character only has four combos. By completing levels, coins and gamepads are earned, medals (bronze, silver, and gold) open up new stages, and new combos are unlocked. New combos can then be swapped and up to four can be equipped at a single time. Certain combos need to be bought in an in-game store after unlocking them, either with coins or gamepads. Initially, three characters are available, each with their own set of combos: Gina, Lester, and Parker. Two more can be unlocked with gamepads: Dolph and Sammo. The remaining ones need to be bought with real money as an in-app purchase: Joe (from Viewtiful Joe), and Blanka, Chun-Li, and Ken (from Street Fighter).

When defeated you can call in a real life friend to rescue you. When he or she completes the round with their character, your initial character is healed and the friend's score is converted into life points for you, with additional ones converted into score points. This can only be done once per game. Friends are managed through a separate crew screen.

The game has a store where boosters (bonuses for the Combo Crew mode), perks (permanent upgrades), skins, and packs can be bought.
