In Los Aliens, the player's objective is solving 170 handcrafted puzzle levels. Within levels, the player controls an alien unit (a spaceship crew member) that can jump in an L-shaped pattern, just like a knight in chess. The goal is to complete all level objectives within a limited amount of moves or time (in seconds). Level objectives usually consist of "complete X lines" and (often multiple) "collect Y special tiles" on a square tile matrix. The plot is that the alien travels through the galaxies and needs to refill its ship's fuel by completing puzzles / levels and recruiting new crew members.

Lines are completed by jumping on all tiles of a matrix' row or column. Upon line completion, all tiles on that line are emptied and all columns right to / rows below the completed line are moved to the left / upwards with then empty lines being refilled at random with new special, empty, or already populated tiles. Special tiles affect gameplay and include, among others, options like tile teleporters, bombs, power-ups (like additional moves), aliens (e.g. that move in a pattern and swap the tile they step on with the one they were previously on). Available crew members used in missions are unlocked through gameplay and vary slightly in character stats (e.g. additional moves or additional time in time-limited missions).
